Taq DNA polymerase
Recombinant Taq DNA polymerase has 5 ?-3 ? DNA-dependent polymerase activity and 5'-3' exonuclease activity of native Taq DNA polymerase from Thermus aquaticus. The rate of Taq DNA polymerase progression depends on the complexity of the DNA template and is approximately 1.5-2 kb/min.

Taq DNA polymerase is a single polypeptide with a molecular weight of about 95 kDa. The polymerase is highly processive and can amplify fragments up to 10 thousand base pairs long. The enzyme has pronounced 5'-3' exonuclease activity and adenyltransferase activity. Transferase activity results in the addition of a single pA to both 3' ends of the double-stranded DNA obtained during amplification. The enzyme has the activity indicated on the label at 74°C and pH 8.8.
Mg2+ ions are absolutely necessary for the activity of Taq DNA polymerase. The enzyme is most active in the Mg2+ concentration range of 0.5-6 mM.

Taq DNA polymerase was purified from an E. coli strain containing a plasmid with the cloned full-length Thermus aquaticus DNA polymerase gene.
Unit of activity
One unit of activity corresponds to the amount of enzyme required to incorporate 10 nmol of dNTP into the acid-insoluble DNA fraction in 30 min at 74°C.
