Modified M-MuLV reverse transcriptase is an RNA-dependent DNA polymerase. The enzyme is capable of synthesizing a complementary DNA strand using single-stranded RNA or DNA as a template. Genetic engineering modification of M-MuLV RT resulted in complete destruction of RNase H-like activity.
Source
Strain E.coli, carrying a plasmid with cloned M-MuLV RT gene with a deleted fragment providing RNase H-like activity.

One unit of activity corresponds to the amount of enzyme required to incorporate 1 nmol dTTP into the acid-insoluble DNA fraction in 10 min at 37°C, using poly(riboA)-oligo(deoxyT)12 as a primer template.
